[Epub ahead of print] , Word Count: 333 OBJECTIVE: This is a systematic review about the effects of low-intensity pulsed ultrasound (LIPUS) on stem cells differentiation. BACKGROUND DATA: In latest studies, several types of stem cells from different sources in the body have been used. These stem cells should be strictly certified and promoted for cell therapies before medical application. Low intensity pulsed ultrasound (LIPUS) has been extensively used in treatment centers and research to promote stem cells differentiation, function and proliferation. MATERIALS AND METHODS: We searched in PubMed, Google Scholar, Medline, and Scopus databases for abstracts and full text scientific papers from 1989-2019 that reported the applications of LIPUS on stem cell differentiation. Related English language articles were located by the following defined key words: low intensity pulsed ultrasound, stem cell, differentiation. Criteria for inclusion in the review were: LIPUS with frequencies of 1-3 MHz and pulsed ultrasound intensity of <500 mW/cm2. Duration, exposure time, and cell sources were taken into consideration. RESULTS: There were 52articles selected according to the inclusion criteria. Most articles demonstrated that application of LIPUS had positive effects on stem cell differentiation. However, some authors recommended that the combined effects of LIPUS with other physical therapy aides were more effective in the differentiation levels of stem cells. CONCLUSIONS: LIPUS significantly increases the level of stem cell differentiation in cells mainly derived from BM MSCs. Clearly, there is a need for further studies to analyze the effect of LIPUS on cells derived from other sources, particularly ADS for treating hard diseases such as osteoporosis and DFU. Due to lack of reporting on standard LIPUS parameters in the field, there is a need more experiments for comparing the protocols for standardization of LIPUS parameters, in order to establish the best protocol.
